Library / Automated ELISA / Steps

Protocol steps

The full run, executed hands-free and vision-verified at every transfer.

1

Coat

The robot dispenses capture antibody into all 96 wells and starts the coating incubation. Vision confirms full-column coverage before incubating.

2

Wash & block

Three wash cycles with PBST, then blocking buffer across the plate. Aspirations are checked for complete well clearance between cycles.

3

Add samples & standards

Samples and the standard curve are dispensed to their mapped wells with fresh tips per transfer to prevent carryover.

4

Detect

Detection antibody is added, incubated, and followed by another wash series to remove unbound conjugate.

5

Develop & read

Substrate is added and stopped after the timed development, then the plate moves to the reader for absorbance at 450 nm.
